will a 2 inch deep substrate of flourite sand create any cold spots or is 2 inches shallow enough where it will allow the heat to go all the way to the bottom

The temperature of the substrate should be ok. I like to line the bottom of my tanks with insulation board to slow down heat loss through the bottom glass. I believe that the use of substrate heaters is generally regarded as unecessary,

so if i use a piece of styro on the bottom i should be fine?

How cold do your rooms get?

I don't think there is any proof that plants prefer warm feet. At least with normal room temperatures it shouldn't be anything to worry about.
