yes, everytime you do maintenance on the CO2 tank, bubble counter etc, the water will go backwards.

Don't mean to hijack this thread, but does the check valve have to be metal? Will one of those plastic ones suffice (like the ones used on air pumps to prevent water from entering the pump)?

The plastic ones will work. But they fail over time, and if you don't check them regularly, could lead to bad problems w/ a ruined regulator.

I see. Thanks for clearing that up for me Epicfish. I was wondering why most use a metal (brass?) check valve.
