Where does most of the bacteria grow??

On my canister Prefilter? In Canister filter? Gravel? or my Wood?

Helping a friend cycle his tank, and not sure which the best spot to get the most bacteria from.. Should i get the juice from the pre filter? or the juice inside the canister filter?? i use a sponge filter as a pre filter, but i only put it on there last week or so

How big of a tank does your friend have? Depending on how much you give your friend, they may still experience a mini-cycle while the bacteria is populating the new tank. Either way, it shouldn't last nearly as long as a typical cycle.

I'd tell your friend to check the water parameters every day for a week and see how things look and go from there.

is it best for him to put it in his filter, or should we just put it in the tank, probably the filter huh

A cup should be fine to get him going. You don't want to get rid of too much media from your tank or you could run into issues with your tank. I would recommend putting it in the filter.
