DIY Yeast CO2 - possible to have it below the tank level?

I'm about to setup a DIY Yeast CO2 reactor, etc and want to know if I can have the bottle below the tank level on the floor? I will be using a check valve and a diffuser.

Yes. It does not make it harder or easier for the CO2 to come up to the tank. Is all pressure...
