Test water prior to flooding?

So I've given dry start a go, the hairgrass has rooted and spread a bit, I'm happy with it aside from spots mold has taken it, but the moss isn't doing that well in spots and I'm tired of fighting mold. As someone who has never really had plants before, what all should I test my water for prior to flooding? I have an RO unit, but I take it I should use the normal water as it has more minerals? Just looking for a bit of insight before I do harm to any progress? What all should I look for in my water prior to flooding?
