Do you mean using an air pump with the air stone and also a separate diffuser? If so, the air stone will greatly lower the amount of co2 that stays in the water - surface agitation causes the co2 to get released from the water

I'm waiting for my supplies still, but I plan to run the co2 during the day with the lights on and an air wand at night when the lights are off

I'm still pretty new to co2 injection myself but the air stone pumping air in along with the co2 diffuser and co2 injection is something i've heard defeats the purpose of the co2 causing it to be released into the air

You should look into a power head instead of an airstone. Thats what I do. Works very well.
